KAPATaq Hot Start DNA Polymerase
KAPATaq Hot Start is a chemically modified version of the single-subunit Taq DNA polymerase enzyme from the thermophilic bacterium Thermus aquaticus, purified from recombinant Escherichia coli.
KAPATaq Hot Start provides the following advantages:
- Improved PCR specificity
- Improved PCR sensitivity
- Higher yields
- Convenience – assembly of PCR reactions at room temperature
- Short activation time - 5 minutes at 95°C
All thermophilic DNA polymerases used in PCR exhibit a small but measurable activity at room temperature. A number of non-specific priming events occur under the low stringency conditions of ambient temperature. The amplification of these non-specific products consumes PCR reagents and competes with the amplification of the desired specific product.
KAPATaq Hot Start DNA polymerase is inactive at room temperature and is reactivated by a 5 minute incubation step at 95°C. Hot start modification eliminates undesirable amplification products resulting from non-specific primer annealing and enzyme activity at room temperature. Many instances of high background amplification and low yields of specific PCR products can be successfully addressed by using KAPATaq Hot Start.
Product Applications
KAPATaq Hot Start DNA Polymerase is ideally suited for:
- High throughput PCR
- Amplification of low copy DNA templates
- Multiplex PCR
- Specific amplification of complex templates
- RT-PCR
