Shoreline Biome assay kits provide significant improvement over many of the standard protocols that are known to miss microbes or provide biased results. All aspects of the systems are designed from the ground up to work together on the industry leading long read PacBio and short read Illumina sequencing platforms.
Each Shoreline Biome assay kit includes the reagents and barcoded primers needed to easily and quickly lyse samples and prepare amplicons for 16S sequencing.
The assay kits for the Illumina sequencing platform contain one 96-well plate for lysis, one tube of DNA purification beads, and one 96-well PCR plate containing 96 barcoded primers targeting either the V1–V3 or V4 region of the 16S rRNA gene. Following PCR amplification, the amplicons containing the Illumina sequencing adaptor are pooled, purified, and are ready for sequencing.
The assay kits StrainID for the PacBio sequencing platform contain one 96-well plate for lysis, one tube of DNA purification beads, and one 96-well PCR plate containing 96 barcoded primers targeting either the full 16S rRNA gene or the 16S-ITS-23S region. Following PCR amplification, the amplicons are pooled and ready for the PacBio SMRTbell reaction. There are additional 16-sample kits for both the full 16S and the 16S-ITS-23S that can be combined with the corresponding 96-well kit, or run separately.
These kits include a royalty free license of the SBanalyzer software, powered by the Athena reference database. The software enables strain level identification. Together, they are the Shoreline Complete StrainID solution.
For maximum flexibility Shoreline Biome is offering the kit items separately. When you already have your extraction optimized there is a possibility to purchase only the Shoreline Wave amplicon kit. Otherwise if you would like to only optimize your extraction, the Rapid Prep kits will be your solution.

The Shoreline Wave amplicon kits are a convenient solution for microbiome researchers seeking the innovative amplicon technology from Shoreline Biome. The smart primer design and unique dual-end barcoding reduce amplification bias and maximize usable reads. Full length 16S and StrainID (16S-ITS-23S) amplicon kits are available for the PacBio Sequencing platforms in 16 sample and 96 sample formats. Kits for the Illumina platforms are available for V1-V3 and V4 amplicons in 16 sample and 96 sample formats.
The Shoreline Rapid Prep is a fast and easy-to-use kit designed for efficient and unbiased isolation of high molecular weight (HMW) single-stranded DNA (ssDNA). Isolate DNA in excess of 40 kB from Gram-negative bacteria, Gram-positive bacteria, stool samples, and vegetive fungal cell isolates. The ssDNA output is ideal for downstream PCR applications targeting specific bacterial or fungal genes of interest. The patented* lysis/DNA purification protocol ensures that no microbe is left behind without the need for cumbersome bead beating, while preserving DNA integrity.
The Shoreline Biome DNA preparation kit lyses the most difficult microbes that are mostly, or even entirely, missed by standard DNA preparation methods. This ensures that your final results reflect the taxonomic diversity and microbial population ratios in your samples of interest.
Based on your experimental needs and the availability of sequencing platforms, Shoreline Biome kits offer optimized PCR choices for parallel sequencing of over 100 individual samples targeting the 16S rRNA microbial gene. The kits take the guesswork out of which target sequences to choose, what polymerase to use, and the level of primer degeneracy. They are designed and validated with multiplex primers for sequencing many samples simultaneously in a single sequencing run. The PCR conditions were optimized by sequencing standard microbiomes to ensure conditions that result in correct microbial profiles.
Shoreline Biome software, for the PacBio long-read sequencing data, sorts samples by barcode and allows tracking by user-selected sample names. Each sample’s reads are mapped to determine phylogenetic identity. Output can be transferred to a spreadsheet to compare results between individual samples or combine samples into virtual groups for comparison at the phylum, strain, or other taxonomic levels. Mapping allows investigators to quickly test hypotheses about variations in microbial types between microbiome cases and controls. Reads can also be used for custom analysis, as desired.
